Definition

BRAF (v-raf murine sarcoma viral oncogene homolog B1) was first cloned by Sithanandam et al. in 1990. It encodes a protein belonging to the RAF family of serine/threonine protein kinases involved in the transduction of mitogenic signals from the cell membrane to the nucleus. BRAF protein phosphorylates MAP 2K1, and thereby activates the mitogen-activated protein kinase (MAPK) signal transduction pathway which affects cell division, differentiation, and secretion.

In mammalian cells, RAF family members are composed of three conserved domains – conserved regions 1, 2, and 3 (CR1, CR2, and CR3), respectively. CR1 is a RAS GTP-binding self-regulatory domain, CR2 is a serine-rich hinge region, and CR3 is a catalytic serine/threonine protein kinase domain that phosphorylates a consensus sequence on protein substrates.
